Perkins is having a tremendous season as the Twins closer, I think he would be the kind of acquisition the Sox could justify giving up a couple of good prospects for, since he is signed affordably through 2015 (with a 2016 club option). Now the Twins are likely not very motivated to move him because of the club friendly contact 3 yrs - $10.3M, but if the Sox could make a deal for Perkins, he would slot in at closer and allow everyone else to slide down into their natural role. My question would be, can the Sox acquire Perkins with some of the excess 40 man players (ie Britton, Brentz, Hassan etc), would a package of Britton, Brentz and Vitek even be close?

No. Elite relievers on premium contracts cost a lot. Think about what the Red Sox gave up for Melancon and then double it.

Perkins had 18 career saves heading into this season. Why is closer his natural role and not Uehara's? Uehara has a better ERA, K rate, and walk rate than Perkins in more innings.
The Red Sox need to stop trading for closers. Saves are overvalued on the trade market. The Melancon, Bailey, and Hanrahan were all sort of defenisble individually in that they didn't give up anyone who was a major contributor, but on the whole they've essentially traded Lowrie, Melancon, Reddick, Head, Alcantara, Pimentel, DeJesus, and Sands for Bailey, Hanrahan, and Brock Holt. Closers cost too much for the value they provide.
